چکیده
Oxidative stress due to free radicals is involved in the pathogenesis of many chronic disorders including cancer, inflammation and neurological diseases. Reactive oxygen species (ROS) may be implicated in age-related macular degeneration (AMD).This study investigated the expression of manganese superoxide dismutase MnSOD on mRNA and protein level. Moreover it examines the genetic sequence polymorphism. Our intend is to find a correlation between the expression of studied genes and nucleotide sequence polymorphisms encoded in the gene of two different types of age-related macular degeneration. Methods: MnSOD gene polymorphisms were determined by PCR/RFLP method. Real time PCR and ELISA methods were used to analyze gene expression on mRNA and protein level. Results: There are statistically significant differences in genotype distribution between patients with AMD and controls. Results showed positive correlation between MnSOD expression on mRNA and protein level and gene sequence polymorphism. Conclusions: The obtained results suggest a role of genetic MnSOD polymorphism in the development age-related degeneration. Posters

Evaluation of CYP2C9 activity in rats: use of tolbutamide alone and in combined with bupropion
A “cocktail”of several probe drugs is often used to evaluate metabolic activity of multiple cytochrome P450 enzymes in one session. Some interactions among probe drugs can appear and may impact the rate of biotransformation of other ones. Our presented work was to aim on the influence of bupropion on rat cytochrome P450-mediated metabolism of tolbutamide. The biotransformation rates of tolbutam...

Cytochromes P450 (P450s or CYPs) belong to a multigene family of more than 3,000 heme proteins which catalyse the NADPH-dependent monooxygenation and other about 60 distinct classes of biotransformation reactions. Cytochromes P450 are known to be involved in the metabolism of over 1,000,000 different xenobiotic and endobiotic lipophilic substrates (Shumyantseva, Bulko, Archakov, 2005). Cytochro...
متن کاملStudy of Cytochrome P450 1A1 (T3801C) Single Nucleotide Polymorphism in Patients with Breast Cancer in Mazandaran Province-Northern Iran
Background: Breast cancer is the first leading cause of cancer-related death in women. Pesticides which are excessively used in northern Iran are one of the most important risk factors for breast cancer incidence. The cytochrome P450 1A1 (cyp1A1) is a key enzyme in xenobiotics metabolism and SNPs of its coding gene has been verified to be important in cancer susceptibility. The aim of thi...

Cytochrome P450 (P450) is the superfamily of enzymes responsible for biotransformation of endobiotics and xenobiotics. However, their large isoform multiplicity, inducibility, diverse structure, widespread distribution, polymorphic expression, and broad overlapping substrate specificity make it difficult to measure the precise role of each individual P450 to the metabolism of drugs (or carcinog...
